GRANISETRON HYDROCHLORIDE BP

Granisetron hydrochloride is the drug used to treat nausea and vomiting especially with particular types of cancer chemotherapy and radiation.

Physical Properties: White or almost white powder

Solubility: Freely soluble in water, sparingly soluble in methylene chloride

Pharmacokinetics of Granisetron Hydrochloride

Absorption: Mean C max is 64 ng/Ml.

Distribution: Vd is about 3 L/kg and Protein binding is 65%.

Elimination: In normal case, about 11% of the orally administered dose is eliminated unchanged in the urine in 48 hours. The remnants of the dose are excreted as metabolites, 48% in the urine and 38% in the faeces.

Pharmacodynamics of Granisetron Hydrochloride

Granisetron Hydrochloride is a potent anti-emetic and highly choosy antagonist of 5-hydroxytryptamine (5-HT3) receptors. Radioligand binding studies have established that granisetron has negligible affinity for other receptor types including 5-HT and dopamine D2 binding sites.

Granisetron is efficient intravenously, either prophylactically or by intervention, in eliminating the retching and vomiting evoked by administration of cytotoxic drugs or by entire body Xirradiation.

Medical uses of Granisetron Hydrochloride

Granisetron Hydrochloride has following medical use:

  • To prevent from chemotherapy-induced nausea and vomiting
  • To prevent from radiation-induced nausea and vomiting (Only Oral Administration)
  • Postoperative nausea and vomiting (through injection only)
  • Side effects of Granisetron Hydrochloride

    It could have following minor as well as serious side effects:

  • Chest pain
  • Pale skin
  • Feeling like you might pass out
  • Headache
  • Diarrhea or constipation
  • Overdose of Granisetron Hydrochloride

    Though, there is no specific treatment for granisetron hydrochloride overdosage; however, in case of overdose, symptomatic treatment should be given.

    Cautions of Granisetron Hydrochloride

    This drug may cause you dizzy or drowsy so, do not drive, use any machinery, or do not perform any activity that requires alertness until you are confident that you can perform such activities safely. Usually, Granisetron is not always effective for treating delayed nausea and vomiting.
